AOD-9604 is a synthetic 16-amino-acid fragment corresponding to the C-terminal region (residues 176-191) of human growth hormone, modified with an additional stabilizing amino acid. The design rationale, developed at Monash University in the 1990s, was to isolate the region of the GH molecule proposed to drive lipolysis (fat breakdown) and inhibition of lipogenesis (fat storage) while removing the region responsible for GH's growth-promoting and insulin-desensitizing effects, since the fragment does not bind the classical GH receptor in the same way as full-length GH. In early rodent studies it was reported to stimulate hormone-sensitive lipase and inhibit acetyl-CoA carboxylase in adipose tissue, reducing body-weight gain and adipocyte size in obese Zucker rats without inducing insulin resistance or elevating IGF-1.
