Mechanism

GHK-Cu is a naturally occurring copper complex of the human tripeptide GHK (glycyl-L-histidyl-L-lysine), found in plasma, saliva and urine, with levels declining with age. Preclinically, it acts as a broad signaling molecule rather than a single-target drug: it delivers copper needed as a cofactor for antioxidant enzymes (e.g., superoxide dismutase), modulates the balance of matrix metalloproteinases and their inhibitors to support extracellular-matrix remodeling, stimulates fibroblast collagen, elastin, and glycosaminoglycan production, and has been reported in gene-expression screens to shift the activity of thousands of genes toward patterns associated with tissue repair and away from patterns associated with inflammation and fibrosis. It also appears to support angiogenesis and hair-follicle stem cell activity in animal/cell models.

GHK-Cu is one of the better-studied cosmetic peptides, with decades of in vitro, animal, and topical human cosmetic-formulation research. Topical human studies (mostly small, industry-linked, or open-label) suggest improvements in skin firmness, fine lines, and photodamage markers with regular cosmetic use. Evidence for injectable/systemic use in humans is far more limited than for topical cosmetic use, and rigorous, independent, large-scale randomized controlled trials for injectable or 'anti-aging' systemic use are lacking. Topical GHK-Cu in approved cosmetic concentrations has a long history of use with a generally favorable safety profile and minimal reported irritation in most formulations. Injectable or high-concentration systemic use is far less studied in humans; copper is a redox-active metal and excess systemic copper exposure carries theoretical oxidative-stress and toxicity concerns, particularly with unregulated dosing or impure product.
