Matrixyl is a synthetic lipopeptide: the pentapeptide KTTKS (Lys-Thr-Thr-Lys-Ser) — a fragment naturally released from the C-terminal propeptide of type I procollagen — conjugated to palmitic acid to improve lipophilicity and penetration across the stratum corneum. KTTKS acts as a matrikine, a peptide 'feedback' signal that the skin interprets as evidence of matrix turnover, cueing dermal fibroblasts to up-regulate synthesis of type I and III collagen, fibronectin and glycosaminoglycans. The palmitoyl tail is a delivery modification and is not itself bioactive. Because it is applied topically at very low concentrations (parts per million) as a cosmetic active, its action is confined to the skin surface and upper dermis; it is not designed for, or studied as, a systemic drug.
