Mechanism

Kisspeptin-10 is the C-terminal decapeptide fragment of kisspeptin, the peptide product of the KISS1 gene and the shortest fragment that retains full receptor activity. It binds the G-protein-coupled receptor KISS1R (formerly GPR54) on hypothalamic neurons that produce gonadotropin-releasing hormone (GnRH). Receptor activation drives GnRH secretion into the hypophyseal portal system, which in turn stimulates pituitary release of luteinizing hormone (LH) and, to a lesser degree, follicle-stimulating hormone (FSH), placing kisspeptin signaling at the top of the hypothalamic-pituitary-gonadal (HPG) axis. Kisspeptin neurons integrate sex-steroid feedback and metabolic signals, and human imaging work suggests kisspoptin also modulates limbic circuits involved in sexual and emotional processing. Its plasma half-life is only a few minutes, so effects are transient.

Evidence

Human physiology studies are substantial for an investigational peptide. Jayasena et al. (2011) showed that kisspeptin-10 stimulates LH release with marked sexual dimorphism and menstrual-cycle-phase dependence. The longer kisspeptin-54 isoform has been tested as an oocyte-maturation 'trigger' in IVF (Abbara et al., 2015), and Mills et al. (2023) reported that kisspeptin modulated sexual brain processing and penile tumescence in men with hypoactive sexual desire disorder. Despite these signals, kisspeptin remains an experimental agent: no formulation is approved for any indication, trials are small and short-term, and long-term safety data are lacking.
